If you cant direct hit then hold till close to target and release. Hold the fire button and release to cause them to detonate. If you direct hit an enemy, it doesn't matter if you are holding the fire button or not. This doesn't apply to wave frame grenade launchers though, which don't have a hold and release mechanism.
